The calculator tells you how many units to draw. Here is exactly what a unit is, and why it means the same thing on every insulin syringe.
On a U-100 insulin syringe one unit is a hundredth of a millilitre. So 10 units is 0.1 mL, and 100 units is 1 mL. Units simply make very small volumes easier to read off the barrel.
It does not matter which insulin syringe you pick up. Drawing to 20 units pulls the same 0.2 mL on a 0.3 mL, a 0.5 mL or a 1 mL syringe. The tick marks never change size.
What changes between syringes is how many of those marks fit on the barrel. A bigger syringe is not a bigger unit, it is just more room.
Example: a 20-unit draw is 0.2 mL on all three syringes above. The only difference is how much empty barrel is left behind it.

Peptide reconstitution calculator math, step by step
The arithmetic behind every peptide reconstitution calculator is the same, and it is worth understanding rather than trusting blindly.
Concentration is mass divided by volume. A 10mg vial reconstituted with 2mL of bacteriostatic water gives 5mg per mL. The volume needed for any given dose is that dose divided by the concentration, so a 2.5mg dose from a 5mg/mL solution takes 0.5mL. Because one unit on a U-100 insulin syringe is 0.01mL, that 0.5mL is 50 units.
Change the water volume and every downstream figure changes with it. The same 10mg vial reconstituted with 5mL gives 2mg per mL, and the same 2.5mg dose now sits at 125 units, which is more than a 1mL syringe holds. Nothing about the compound changed. Only the dilution did.

Using the peptide stack calculator for blends
Blended vials such as GLOW 70 and KLOW 80 state a total mass across several compounds, and those compounds are not present in equal shares. Running a peptide stack calculator means treating each component separately: take its individual mass from the product documentation, then calculate its own concentration against the same water volume. One vial produces several concentrations, not one.

The calculation survives about as long as your memory of it, which is not long. Record the water volume and the resulting concentration on the vial at the moment of reconstitution, alongside the date of first puncture and the lot number. Once liquid has been drawn, the original volume cannot be reconstructed by looking, and every figure calculated afterwards is a guess. The most common question typed into any peptide calculator is some version of how much bac water for 10mg retatrutide. There is no single correct answer, because the water volume is a choice, not a property of the compound. What the volume determines is the concentration, and the concentration determines what the syringe reads.
Run a 10mg retatrutide vial at 1mL and the solution is 10mg/mL, so 1mg sits at 10 units. Run the same vial at 2mL and it is 5mg/mL, putting 1mg at 20 units. At 3mL it is 3.33mg/mL and 1mg reads 30 units. Larger volumes give finer resolution on the syringe and shorter in-use windows once the vial is punctured. That trade-off is the entire decision.

They all describe the same two divisions: mass over volume gives concentration, dose over concentration gives volume.
Anything claiming to do more than that is either adding a syringe conversion, which is a third division by 0.01, or making a recommendation, which is not arithmetic and not something a calculator is qualified to do.

It will not tell you how much of anything to use. Every figure on this page is downstream of numbers you type in, and the tool has no opinion about whether those numbers are sensible.
