Retatrutide vs Semaglutide: What the Trial Data Compares

Retatrutide vs semaglutide, and retatrutide vs Ozempic, is a comparison between a triple receptor agonist and a selective GLP-1 receptor agonist, and the numbers people quote for each come from different trials. Semaglutide acts on the GLP-1 receptor. Retatrutide acts on three: GIP, GLP-1 and glucagon. Each has a large published trial in adults with obesity, and no head-to-head between the two appears in the papers cited here. This page lays out what each trial measured, why the headline percentages can’t be lined up cleanly, and what a batch lab report shows for the research vials. Everything below describes laboratory research and published trials, and nothing here is medical advice.

Retatrutide vs semaglutide: what is the difference?

Semaglutide is a selective GLP-1 receptor agonist, a description used in the SURPASS-2 paper. Retatrutide is a triple agonist of the GIP, GLP-1 and glucagon receptors. That is the mechanistic difference: two extra receptors, one of which, glucagon, has a different biology from the incretin pair. The research difference is maturity. Semaglutide’s obesity trial program reported first and has a long track record. Retatrutide’s phase 3 obesity results have now been published too, which changes what an honest comparison can say, and it also means searches for retatrutide vs Ozempic are really searches for retatrutide against semaglutide.

What the retatrutide trials measured

The first dose-ranging result is Jastreboff and colleagues’ phase 2 trial in the New England Journal of Medicine: 338 adults over 48 weeks, with mean weight change of -24.2% in the highest arm against -2.1% for placebo. Gastrointestinal events were the most common adverse events, and heart rate rose in a dose-dependent way, peaking at 24 weeks and declining after. The phase 3 result is the TRIUMPH-1 trial in the same journal, in 2,339 adults with obesity and no diabetes over 80 weeks, with weight change of -25.0% at the highest arm against -3.9% placebo, and improvements in knee osteoarthritis pain and sleep apnea events in the subgroups that had them. In type 2 diabetes, a phase 3 monotherapy trial in The Lancet of 537 adults reported HbA1c falling by 1.94% at the highest arm against 0.81% for placebo over 40 weeks. The what retatrutide does page covers the mechanism in plain language.

What the semaglutide trials measured

Wilding and colleagues’ STEP 1 trial in the New England Journal of Medicine randomized 1,961 adults with overweight or obesity, without diabetes, to semaglutide or placebo plus lifestyle intervention for 68 weeks. The change in body weight was -15.3 kg against -2.6 kg, and 50.5 percent of the semaglutide group reached a 15 percent loss or more, against 4.9 percent on placebo. Nausea and diarrhea were the most common adverse events, and gastrointestinal events led to discontinuation in 4.5 percent against 0.8 percent on placebo. In type 2 diabetes, semaglutide was the comparator in SURPASS-2, which is covered on the semaglutide vs tirzepatide page.

Why the retatrutide and semaglutide numbers don’t line up

Three things stop a clean subtraction. First, duration: 80 weeks against 68 weeks, which gives weight more time to change in one than the other. Second, populations and designs: different eligibility rules, different sponsors, different lifestyle components and different dose-escalation schedules. Third, the way results are reported, with percentage change in one and kilograms in the other, and different estimands in each. A rough reading is that retatrutide’s top arm produced larger average weight loss than semaglutide’s trial did, and that is all the data supports. A head-to-head, which is how the tirzepatide and semaglutide question was answered, hasn’t been published for this pair in the sources cited here.

Who was in the retatrutide and semaglutide trials

The populations matter as much as the percentages. STEP 1 enrolled 1,961 adults with a body-mass index of 30 or more, or 27 or more with a weight-related condition, and no diabetes. TRIUMPH-1 enrolled 2,339 adults with obesity and no diabetes, with prespecified subgroups of people who also had knee osteoarthritis or obstructive sleep apnea, so it measured pain and apnea events as well as weight. The retatrutide type 2 diabetes trial is a separate population altogether, adults with diabetes inadequately controlled by diet and exercise, and its endpoint was HbA1c. A figure from one population can’t be transplanted to another, and the sections above keep the three records apart for that reason.

Reading headlines about retatrutide vs Ozempic

Headlines tend to quote the biggest single number from each side and set them next to each other. A fair reading asks four things of each figure: which trial, how long, which dose arm, and which population. Quoting -25.0% beside a semaglutide figure answers none of them. The trial abstracts cited here are written to report those details, which makes them a better source than a summary. When a page leaves them out, it is usually because the comparison looks cleaner without them.

Does the glucagon receptor make retatrutide better?

The extra receptors are why retatrutide is studied, not proof that it wins. Glucagon receptor activity is a different lever from the incretin pair, and the trials show large weight changes alongside dose-related heart rate increases in phase 2. Whether the third receptor accounts for the extra weight loss, or the GIP receptor does, or the dose range does, can’t be separated by comparing two trials. That would take a design with matched arms.

Retatrutide vs semaglutide in women’s research

Women are well represented in some of these trials: 55 percent of the retatrutide type 2 diabetes trial’s participants were female, while the phase 2 obesity trial enrolled 51.8 percent men. The abstracts for the large obesity trials don’t give sex-stratified outcomes. A woman framing her own research question will want those breakdowns from the full papers. The peptides for weight loss in women guide covers that setting, and semaglutide for women goes through the older compound.

What the retatrutide and semaglutide studies can’t tell you

Trials of the pharmaceutical products can’t be transferred to research-grade vials, and nothing here is guidance for any use. The trial arms used specific titration schedules under clinical supervision. Gastrointestinal effects were the most common adverse events for both compounds, and the long-term picture for retatrutide is still being written. Industry sponsors funded the trials, which is normal and worth knowing. For regulatory status of either compound, the regulator is the source, and this page doesn’t state one.

Retatrutide and semaglutide COA: what the lab reports show

Every Healio Peptides batch is tested by an independent US laboratory before release, and the report is published before you order. These are the current two.

Retatrutide Semaglutide
Batch HEALIO-RETA-2602 HEALIO-SEMA10-2607
Purity (HPLC) 99.3% 99.4%
Identity (ESI-MS) Confirmed, conforms to the reference standard (4731.33) Confirmed, conforms to the reference standard (4113.58)
Endotoxins (LAL) Under 0.5 EU/mg (limit 5 EU/mg) Under 0.5 EU/mg (limit 5 EU/mg)

Both are long peptides, so identity is the row to read closely. Purity says how clean the powder is, and identity says it holds the molecule on the label. Match the batch and lot numbers to the vial. Reading a peptide certificate of analysis explains every row.

Retatrutide vs semaglutide price and format

Healio Peptides lists retatrutide in 10mg and 20mg and 30mg vials and semaglutide in 5mg and 10mg vials, both lyophilized powder. Strength differs between vials, so compare cost per milligram and not per vial: retatrutide’s lowest listing is $7.90 per mg and semaglutide’s is $16.00 per mg. Both reconstitute with bacteriostatic water for sale, and the retatrutide calculator and semaglutide calculator turn vial strength and water volume into a concentration. How to reconstitute peptides covers the handling.

Where to buy retatrutide and semaglutide for research

Whichever supplier you use, look for a published certificate before anything else. Every Healio Peptides batch ships with a third-party report, and orders go out from the United States. Start with the retatrutide for sale and semaglutide for sale pages, both of which carry the certificate before you order. The peptides for weight loss category shows the rest of the range.

Retatrutide vs semaglutide: frequently asked questions

What is the difference between retatrutide and semaglutide?

Retatrutide activates three receptors, GIP, GLP-1 and glucagon. Semaglutide is a selective GLP-1 receptor agonist.

Is retatrutide stronger than semaglutide?

The trials point that way: -25.0% at the top arm of TRIUMPH-1 against a semaglutide trial result of -15.3 kg. They were different trials, though, and no head-to-head is in the papers cited here, so it isn’t a like-for-like result.

What does retatrutide vs Ozempic mean?

Ozempic is a semaglutide product, so the search is the same comparison as retatrutide vs semaglutide.

Does the glucagon receptor make retatrutide better?

The extra receptor is the reason retatrutide is studied. Comparing two separate trials can’t show which receptor accounts for a difference in weight change.

Is retatrutide approved like semaglutide?

Regulatory status is the regulator’s to state, and this page cites published trials only. Healio Peptides sells both compounds strictly for laboratory research, and nothing on this page is medical advice.

Do retatrutide and semaglutide reviews tell me anything useful?

Reviews mix different products, sources and expectations, so they say little about any one vial. A batch certificate says more: it ties a purity and identity result to a lot number you can check.
