Understanding units

The calculator tells you how many units to draw. Here is exactly what a unit is, and why it means the same thing on every insulin syringe.

1 1 unit = 0.01 mL

On a U-100 insulin syringe one unit is a hundredth of a millilitre. So 10 units is 0.1 mL, and 100 units is 1 mL. Units simply make very small volumes easier to read off the barrel.

2 A unit is always a unit

It does not matter which insulin syringe you pick up. Drawing to 20 units pulls the same 0.2 mL on a 0.3 mL, a 0.5 mL or a 1 mL syringe. The tick marks never change size.

3 Syringes hold different totals

What changes between syringes is how many of those marks fit on the barrel. A bigger syringe is not a bigger unit, it is just more room.

0.3 mL30 units max
0.5 mL50 units max
1.0 mL100 units max

Example: a 20-unit draw is 0.2 mL on all three syringes above. The only difference is how much empty barrel is left behind it.

How the semaglutide calculator works

Semaglutide is a GLP-1 receptor agonist supplied as a lyophilized powder, and this semaglutide reconstitution calculator does the arithmetic that comes after the water goes in. It opens on a 5mg vial with 2mL of bacteriostatic water, with a 10mg preset one click away. Change the vial, the water or the syringe and the concentration, the volume and the syringe reading all follow. The amount field is left empty, since the amount is set by the protocol a researcher is working to.

Semaglutide reconstitution: why small amounts call for more water

Semaglutide amounts are often discussed in micrograms, and a vial holds milligrams. That gap is why the water volume matters more here than for most compounds. The mcg per unit shortcut makes it visible: concentration in mg/mL multiplied by 10 gives the micrograms in each syringe unit.

  • 5mg vial in 1mL gives 5mg/mL, which is 50mcg per unit
  • 5mg vial in 2mL gives 2.5mg/mL, which is 25mcg per unit
  • 5mg vial in 5mL gives 1mg/mL, which is 10mcg per unit
  • 10mg vial in 4mL gives 2.5mg/mL, which is 25mcg per unit

The fewer micrograms each unit represents, the finer the resolution on the barrel. A stronger solution packs more into each tick and leaves less room for a misread.

Semaglutide dosage calculator: when the syringe is too small

Volume is the amount divided by the concentration, and units are volume times 100. On the 5mg vial in 2mL, 1mg is 0.4mL, which is 40 units. That is more than the 30 units a 0.3mL syringe holds, and the calculator will say so and suggest a larger syringe or less water. Change the syringe size in the form to see how the fill bar responds. The doses-in-vial figure divides the vial by the amount: five 1mg draws from the 5mg vial.

Verify the vial and record the mix

Every figure starts from the strength printed on the vial, so confirm it against the certificate of analysis for your batch. Healio publishes each one on the research page, and how to read a certificate of analysis explains which figures actually prove something. Once the vial is mixed, write the water volume, the concentration and the date on the label. Our guide to reconstituting peptides covers the handling, and how long bacteriostatic water lasts covers the in-use window.

Semaglutide for sale from Healio is stocked in 5mg and 10mg vials. Liraglutide vs semaglutide covers how the two compare. For another incretin-class compound, the retatrutide calculator works the same way, and the all-peptide calculator takes any vial strength.

Semaglutide calculator FAQ

How much bacteriostatic water goes in a 5mg semaglutide vial?

There is no one correct volume. 1mL gives 5mg/mL, 2mL gives 2.5mg/mL and 5mL gives 1mg/mL. Pick the volume that puts your target amount at a comfortable, readable point on the syringe scale.

How do I convert semaglutide mg to mcg?

Multiply milligrams by 1,000. A 5mg vial holds 5,000mcg. In the calculator, switch the dose unit between mg and mcg and the other figures convert automatically.

What does mcg per unit mean?

It is the micrograms in one tick of a U-100 syringe. Multiply the concentration in mg/mL by 10. At 2.5mg/mL that is 25mcg per unit. It is a quick way to sanity-check any reading.

Why does the calculator say the volume is too large for my syringe?

A 0.3mL syringe holds 30 units, a 0.5mL holds 50 and a 1mL holds 100. If the amount needs more units than the syringe holds, use a larger syringe or reconstitute with less water.

Does water volume change how much semaglutide is in the vial?

No. The mass in the vial is fixed. The water volume only changes how concentrated the solution is, which changes how much liquid carries a given amount.

Does the semaglutide calculator say how much to use?

No. It converts the figures you enter and makes no recommendation. Healio supplies research compounds for laboratory use, so quantities belong to the protocol a researcher is working to.

Healio Peptides supplies research compounds intended for laboratory and research use only. This calculator performs concentration arithmetic on figures you enter. It does not recommend quantities, and nothing on this page is medical advice. These compounds are not intended for human consumption and these statements have not been evaluated by the FDA.
